DNA Methylation

DNA methylation is a chemical marking process in which methyl groups are added to DNA, often influencing how genomic regions are regulated.

In humans, methylation is commonly measured at cytosine bases next to guanine, called CpG sites. The underlying DNA sequence stays the same; the chemical mark is part of the epigenetic layer that helps cells regulate gene activity and maintain identity. Patterns vary by tissue, cell type, development, and other biological contexts.

Laboratories can measure selected sites or survey a much larger set. An epigenetic clock applies a mathematical model to a particular subset or combination of methylation values. Sample collection, the mix of cell types in the specimen, laboratory platform, normalization, and algorithm version can influence the resulting estimate.

A methylation association does not by itself establish cause, diagnosis, or a specific action. Many sites have no simple one-to-one meaning, and a clock score compresses a large pattern into a smaller output. Keeping raw measurement, computational model, and interpretation separate makes the result easier to understand.

Methylation can differ between cell populations in the same specimen. Computational adjustment may estimate or account for cell composition, but that adds another model layer. Two tests can therefore examine overlapping biology while producing values that are not directly comparable.

Array-based reports may express methylation at a site as a beta value representing the proportion of measured signal attributed to the methylated state. Quality filtering and normalization occur before those site-level values are used in a clock or other model.
